pjsua_conf_adjust_rx_level will mute the speaker volume.
If you want to mute the mic you can use
pjsua_conf_adjust_tx_level or you can do this
pjsua_call_info callInfo;
pjsua_call_get_info(identifier, &callInfo);
pj_status_t status = pjsua_conf_disconnect(0, callInfo.conf_slot);
where identifier is the call identifier given to you by pjsip.
Also you can mute the mic calling the HW directly
